Live-cell emphasis#

Nikon Ti2-E with a Yokogawa CSU-X1 spinning disk confocal unit

A motorized Nikon Ti2-E body is matched with a Yokogawa CSU-X1 spinning disk confocal unit for fast and gentle live-cell imaging with a 10x7mm field of view. A Cairn TwinCam unit allows for simultaneous dual-color imaging on two Hamamatsu Fusion BTs (sCMOS cameras with a max QE of 95%), and a piezo stage enables rapid volumetric acquisitions. Four laser lines (405, 488, 561, 640 nm) facilitate flexibility in fluorophores. A stage-top incubation chamber provides a cell-friendly environment of 37°C and 5% CO2, and Nikon’s Perfect Focus System enables long-term imaging without drift. Throughput is increased by imaging multiple locations in between time points. A galvo miniscanner allows for selective photomanipulation and enables FRAP, photocaging, photoactivation, etc.

A Hamamatsu Fusion BT (sCMOS camera with a max QE of 95% with an ultra-quiet low read noise mode), and a piezo stage enables rapid volumetric acquisitions with a 10x7mm field of view. Tiling + stitching is possible with flexible region selection options. Four laser lines (405, 488, 561, 640 nm) facilitate flexibility in fluorophores. Six objectives--from a dry 4x to an oil 100x/1.49 NA Apo TIRF--permit imaging at all scales.
